Topic: A method to extract the minimum free-energy path using cryo-electron microscopy images
Abstract: Recently, we developed the cryo-BIFE method, cryo-EM Bayesian Inference of Free-Energy profiles, which uses a path collective variable to extract free-energy profiles and their uncertainties from cryo-electron microscopy images (Giraldo-Barreto, J., Ortiz, S., Thiede, E.H. et al. A Bayesian approach to extracting free-energy profiles from cryo-electron microscopy experiments. Sci Rep 11, 13657 (2021). https://doi.org/10.1038/s41598-021-92621-1). This work was focused on developing, understanding and validating cryo-BIFE for a predetermined path. However, open questions are: how to generate a conformational path for experimental cases and how to optimize the conformational path? The purpose of my talk is to show some preliminary results on the road to solving these questions.
